Penicillin, discovered by Alexander Fleming, belongs to the beta-lactam class of antibiotics and is a widely used broad-spectrum antibiotic which finds a wide range of clinical applications globally. It is effective in treating infections caused by gram-positive cocci, gram-positive rods, most anaerobes, and gram-negative bacteria, among others. Penicillin can also be administered for the treatment of cellulitis, a bacterial infection affecting the skin and deeper tissues.
Penicillin is a class of antibiotics which originates from Penicillium fungi. The beta-lactam ring present in these fungi serves as the fundamental structure of penicillin, playing a vital role in its antibacterial properties. Penicillin G (benzylpenicillin) is water-soluble, whereas Penicillin V (phenoxymethylpenicillin) is acid-stable in nature. Unlike bacteriostatic antibiotics, penicillin is bactericidal, which means it eliminates bacteria instead of just slowing down their growth.
Fermentation is the process commonly used in the commercial production of penicillin. This process is conducted in a stainless-steel tank reactor with a capacity of 30 to 100 thousand gallons in aseptic conditions. This process consists of two to three initial seed growth phases, followed by a production phase lasting 120 to 200 hours. The carbon distribution during fermentation is approximately 65% for cellular maintenance, 25% for growth, and around 10% for penicillin production. Mini-harvest protocols are commonly used in penicillin fermentation, involving the removal of 20-40% of the fermenter contents and replacing it with a fresh sterile medium. This step can be repeated multiple times to increase the overall penicillin yield per fermenter. Penicillin is then produced into the medium and collected for commercial uses in the medicinal sector.

Here are the key steps for penicillin production:
1. Preparation of the Fermentation Medium:
The medium typically contains corn-steep liquor, lactose, glucose, and mineral salts to support mould growth.
2. Fermentation:
The mould is inoculated into the medium and fermented in large bioreactors for several days at controlled temperatures (20-24°C) and pH (6.5). During fermentation, the mould secretes penicillin into the broth.
C6H12O6 (glucose) → Penicillin + CO2 + H2O
3. Extraction:
Penicillin is extracted from the broth using solvents like amyl acetate or butyl acetate, then extracted again into an aqueous solution.
Penicillin + Solvent → Penicillin-Solvent Complex
4. Purification:
The aqueous penicillin is treated with activated charcoal and filtered to remove impurities.
5. Conversion to Penicillin Salts:
The purified penicillin is converted into various salts (e.g., sodium or potassium penicillin) by reacting with appropriate bases.
Penicillin + NaOH → Sodium Penicillin + H2O
6. Final Processing:
The penicillin salts are crystallised, dried, and packaged for commercial use.
Natural penicillin’s find growing applications for the treatment of infections such as strep throat, syphilis, and Lyme disease. On the other hand, nafcillin, oxacillin, and dicloxacillin are classified as penicillinase-resistant penicillin’s, which are available in both intravenous and oral forms and are commonly used in the treatment of staph infections. For instance, Amoxicillin is one of the most prescribed penicillin medications in the world, particularly for the treatment of ear infections, urinary tract infections, and pneumonia, among other diseases. These use cases of penicillin is driving market’s growth.
