Fenofibrate is an oral medication belonging to the fibrate class, primarily used to manage abnormal blood lipid levels, specifically lowering triglycerides and increasing high-density lipoprotein (HDL) cholesterol. Originally named procetofen, it was later renamed to comply with World Health Organisation naming guidelines. The drug was developed by Groupe Fournier SA of France.

Fenofibrate is a white crystalline solid with a melting point of 80.5°C. It has a chemical formula of C20H21ClO4 and an average molecular mass of 360.83 g/mol. The drug is poorly soluble in water but soluble in organic solvents, which aids in its absorption in the gastrointestinal tract. Fenofibrate primarily functions as an antilipemic agent and its mechanism of action involves activating peroxisome proliferator-activated receptor alpha (PPARα), enhancing lipid metabolism and reducing atherogenic particles in the bloodstream.

The production process of fenofibrate begins with the reaction of starting materials, such as clofibrate derivatives, under controlled conditions. Step 1: Synthesis of 4-Chlorobenzhydrol

The first step involves the synthesis of 4-chlorobenzhydrol. 

The chemical reaction is as follows: C6H5CHO + C6H4ClMgBr → C13H11ClO

In this reaction, benzaldehyde (C6H5CHO) reacts with 4-chlorophenylmagnesium bromide (C6H4ClMgBr) to form 4-chlorobenzhydrol (C13H11ClO).

Step 2: Synthesis of 4-Chlorobenzhydryl Chloride

The next step is the synthesis of 4-chlorobenzhydryl chloride. 

The chemical reaction is as follows: C13H11ClO + SOCl2 → C13H10Cl2 + SO2 + HCl

In this reaction, 4-chlorobenzhydrol (C13H11ClO) reacts with thionyl chloride (SOCl2) to form 4-chlorobenzhydryl chloride (C13H10Cl2).

Step 3: Synthesis of Fenofibrate

The final step involves the synthesis of fenofibrate by reacting 4-chlorobenzhydryl chloride with isobutyric acid. 

The chemical reaction is as follows: C13H10Cl2 + C4H8O2 → C20H21ClO4

In this reaction, 4-chlorobenzhydryl chloride (C13H10Cl2) reacts with isobutyric acid (C4H8O2) to form fenofibrate (C20H21ClO4).
