About Etodolac (Lodine)

Etodolac (Lodine) is a nonsteroidal anti-inflammatory drug (NSAID) used to treat osteoarthritis, rheumatoid arthritis, and acute pain. It works by inhibiting the production of prostaglandins, which are substances that promote inflammation and pain. Commonly prescribed dosages include 300 mg and 400 mg, taken either once or twice daily, depending on the severity of the condition. Etodolac was patented in 1971 and approved for medical use in 1985. It has since been manufactured by various pharmaceutical companies, including Forest Laboratories and Teva Pharmaceuticals, under different brand names, with generic etodolac also available.

Properties of Etodolac (Lodine)

Etodolac exhibits analgesic (pain-relieving) and antipyretic (fever-reducing) properties by inhibiting the synthesis of prostaglandins, which play a crucial role in inflammation and pain perception. Etodolac is a white crystalline solid with a molecular formula of C17H21NO3 and a molecular weight of 287.35 g/mol. It has a melting point range of 145-148 °C and is insoluble in water but soluble in alcohols and chloroform.

Manufacturing Process of Etodolac (Lodine)

The production process of etodolac (Lodine) begins with the preparation of raw materials, which involves sourcing the necessary chemical precursors. The synthesis of etodolac includes several steps, starting with the formation of the pyranocarboxylic acid structure, followed by racemisation to obtain a racemic mixture of etodolac. After synthesis, the compound undergoes purification through crystallisation and filtration to isolate pure etodolac. Next, the purified etodolac is formulated by mixing it with excipients such as fillers and binders. The formulation is then processed into tablets or capsules through compression or encapsulation. Quality control is a critical step, where the product is tested for potency, purity, and consistency. Finally, the finished product is packaged for distribution, ensuring that it meets the required standards for safety and efficacy.

Synthesis of Etodolac (Lodine)

The production of etodolac involves several key steps, such as:

Raw Materials:

  • Indole (C8H7N)
  • Ethyl acetoacetate (C6H10O3)
  • Sodium ethoxide (C2H5ONa)
  • Ethyl iodide (C2H5I)
  • Sodium hydroxide (NaOH)
  • Acetic anhydride (C4H6O3)
  • Pyridine (C5H5N)

Process Description:

Step 1: Alkylation of Indole

Indole is first alkylated with ethyl iodide in the presence of sodium ethoxide to form 1-ethylindole.

C8H7N + C2H5I + C2H5ONa → C10H11N + NaI + C2H5OH

Indole + Ethyl iodide + Sodium ethoxide → 1-Ethylindole + Sodium iodide + Ethanol

Step 2: Condensation with Ethyl Acetoacetate

1-Ethylindole is then condensed with ethyl acetoacetate in the presence of sodium ethoxide to form an intermediate product.

C10H11N + C6H10O3 + C2H5ONa → Intermediate + C2H5OH + NaOH

1-Ethylindole + Ethyl acetoacetate + Sodium ethoxide → Intermediate + Ethanol + Sodium hydroxide

Step 3: Cyclisation

The intermediate product undergoes cyclisation to form the core structure of etodolac.

Intermediate → Cyclised Intermediate

Step 4: Acetylation

The cyclised intermediate is then acetylated using acetic anhydride in the presence of pyridine to form etodolac.

Cyclised Intermediate + C4H6O3 → Etodolac (C17H21NO3) + C2H4O2

Cyclised Intermediate + Acetic anhydride → Etodolac + Acetic acid

Step 5: Purification

The crude etodolac is purified through recrystallisation to obtain the final product in pure form.

Etodolac (crude) → Etodolac (pure)

Applications and Drivers of Etodolac (Lodine)

Lodine is mainly used to treat osteoarthritis and rheumatoid arthritis by reducing inflammation and pain. Etodolac is also approved for treating short-term pain and juvenile rheumatoid arthritis. The extended-release form of etodolac is used to manage acute pain, while the immediate-release form is used for long-term joint pain and inflammation in various types of arthritis. These applications make etodolac an important medication for managing arthritis-related symptoms and improving patients' quality of life, driving its demand.
