About Blood Collection Tube

Blood collection tubes, also known as vacutainers, are sterile containers used to collect and store blood samples for various medical tests. They come in different colours, each representing a specific additive or preservative, such as EDTA for haematology tests, sodium citrate for coagulation studies, serum separator tubes for chemistry analyses, heparin tubes for collecting heparinised plasma, and fluoride/oxalate tubes for preserving glucose in whole blood. These tubes ensure the integrity of the blood sample and prevent contamination during transportation and storage. However, proper labelling and handling of blood collection tubes are crucial to ensure accurate test results.

Properties of Blood Collection Tube

Blood collection tubes come in various sizes and colours indicating the additives they contain, such as anticoagulants (EDTA, sodium citrate, heparin) to prevent clotting, clot activators to promote clotting, or gels to separate serum or plasma from cells after centrifugation. The tubes have a vacuum seal and rubber stopper that is punctured by a double-ended needle during blood draw, allowing a predetermined volume of blood to be collected. The order of draw is standardised to avoid cross-contamination of additives between tubes.

Manufacturing Process of Blood Collection Tube

The manufacturing process of blood collection tubes begins with the combination of the rubber stopper cap and the plastic stopper cap using a special cap combination machine. The tubes are then labelled and then an automated dosing machine is used to feed the equipment with the necessary liquid additives for the tubes. The tubes are then capped using capping machines that apply the plastic or metal caps onto the filled tubes. In some cases, the tubes may undergo centrifugation to help spread any additives evenly throughout the tube. The packaged tubes are then placed into a shrink packaging machine that uses a heated air system to shrink a tray of blood collection tubes. Finally, the packaged tubes are placed into cartons for final storage and distribution.

Industrial Manufacturing of Blood Collection Tubes

Step 1: Manufacturing the Glass or Plastic Tubes

1. Starting Materials:

  • Glass Tubes: Borosilicate glass (SiO2, B2O3, Na2O, Al2O3)
  • Plastic Tubes: Polyethylene terephthalate (PET): (C10H8O4)n

2. Chemical Reactions (For PET Production):

  • The synthesis of PET involves the esterification reaction of terephthalic acid (C8H6O4) with ethylene glycol (C2H6O2) to form bis(2-hydroxyethyl) terephthalate.
  • C8H6O4 + 2 C2H6O2 → C12H14O6 + 2 H2O
  • The bis(2-hydroxyethyl) terephthalate undergoes polycondensation to form PET.
  • n C12H14O6 → (C10H8O4) n + 2n H2O

Step 2: Coating the Interior Surface

1. Siliconisation of Glass Tubes:

  • A thin layer of silicone oil (dimethylpolysiloxane, PDMS) is applied to the inner surface of glass tubes to reduce friction and prevent blood cells from adhering.
  • (CH3)3SiO [Si(CH3)2O]nSi(CH3)3

2. Plastic Tubes:

  • The inner surface of plastic tubes is also treated with silicone oil or other surfactants to ensure proper blood flow and reduce cell adhesion.

Step 3: Adding Additives and Anticoagulants

1. Common Additives:

  • EDTA (Ethylenediaminetetraacetic acid): C10H16N2O8
  • Heparin: Complex polysaccharide
  • Sodium Citrate: Na3C6H5O7
  • Potassium Oxalate: K2C2O4

2. Chemical Reactions:

EDTA Addition:

  • EDTA is added to the tubes in its disodium or tripotassium salt form to chelate calcium ions and prevent blood clotting.
  • Ca2+ + C10H14N2Na2O8 → Ca(C10H12N2O8)2- + 2 Na+

Heparin Coating:

  • Heparin is coated inside the tube to inhibit thrombin and prevent clotting.

Sodium Citrate Addition:

  • Sodium citrate is added to chelate calcium and prevent coagulation.
  • Ca2+ + Na3C6H5O7 → CaC6H5O7 + 3 Na+

Potassium Oxalate Addition:

  • Potassium oxalate is added to precipitate calcium and prevent clotting.
  • Ca2+ + K2C2O4 → CaC2O4 + 2 K+

Step 4: Assembly and Sterilisation

1. Tube Assembly:

The tubes are assembled with stoppers and labels. The stoppers are usually made of rubber or plastic and may contain a vacuum to facilitate blood draw.

2. Sterilisation:

The assembled blood collection tubes are sterilised using gamma irradiation or ethylene oxide (EtO) gas to ensure they are free from contaminants.

Step 5: Final Product

The final blood collection tubes are sterile, coated with anticoagulants, and ready for use in medical settings for blood sample collection.

Applications and Drivers of Blood Collection Tube

The blood collection tubes market has a wide range of applications in the healthcare industry. These tubes are primarily used for collecting blood, urine, and serum samples from patients to diagnose various blood-related diseases and perform haematology procedures, such as complete blood count (CBC) tests. They are also used for collecting samples for chemistry, serology, immunology, coagulation, glucose, blood alcohol, and lactic acid tests.

The increase in the number of surgeries and the need for blood transfusions have encouraged the adoption of blood storage practices, which in turn, has fuelled the demand for these tubes. Additionally, the rising prevalence of chronic diseases and advancements in healthcare diagnostics have contributed to the growth of the market.
